Rhapsody Single-Cell Analysis System Configuration:

  • Type: microwell-based, high-throughput single-cell analysis system.
  • Primary Function: capture of multiomic information (gene expression, protein expression, TCR/BCR repertoire) from single cells for next-generation sequencing (NGS) library generation.
  • Cell Throughput per Cartridge: 100 to 40,000 cells.
  • Capture Rate: up to 80% for certain cell types.
  • Specific rates: 40,000 cells: 83% capture, 88% (20k), 90% (10k)
  • Multiplet Rate: 2–3% at 10,000 cell load; 8–10% at 40,000 cell load
  • Microwell Technology: 220,000 microwell partitions per cartridge
  • Loading Volume: 575 uL cell suspension.
  • Gentle Gravity-based Capture: suitable for fragile cell types: granulocytes, neutrophils, CAR-T cells, stem cells, tumor xenograft-derived cells, T cells, NK cells, etc.

Testing performed by our in-house service team:

The Rhapsody Single-Cell Analysis System passed all tests and was found to be fully functional:

  • We powered on and initialized the system.
  • We verified the functionality of the touch screen display.
  • We completed the cell loading process.
  • We completed the bead loading process.
  • We performed and verified the bead wash procedure.
  • We completed the sample retrieval without issues.
  • We tested and confirmed the internal camera function. (Images shown were taken during testing.)
  • We completed motion control testing.
  • We tested tray ejection and retraction.
  • We checked the side door operation.

Instrument Overview:

  • BD Rhapsody Express System uses microwell tech for single-cell capture without fluidic pumps.
  • Each cartridge holds over 220,000 microwells, accommodating up to 40,000 cells.
  • Gravity-based cell settling minimizes stress, ideal for fragile cells.
  • BD Rhapsody Scanner offers visual QC, assessing cell capture and multiplet rates.
  • The system ensures minimal batch effects across replicates and sites.
  • BD Rhapsody Express System supports flexible experimental designs with archived beads.

Instrument Application:

  • Profiling gene expression at the single-cell level
  • Investigating immune cell heterogeneity
  • Identifying rare cell populations
  • Studying tumor microenvironments
